's crizanlizumab has suffered a setback in the UK healthcare market, failing to gain approval from the NATIONAL Institute for Health and Clinical Excellence (NICE), British media reported. Within The scope of Adakveo's listing license, cost watchdog NICE does not recommend that the drug be used to prevent relapsed sickle cell crises (or vascular obstructive crisis, VOCs) in patients aged 16 or older with sickle cell disease.Clinical evidence suggests that patients taking Adakveo had sickle cell crises less often in a year than those who did not receive any treatment. However, according to NICE' research, these results are uncertain because the trial is short-lived and includes only a small number of people who take the licensed dose of the drug.In addition, the cost watchdog noted that there was uncertainty about the cost-effectiveness of the drug, which could be significantly higher than the average cost-effectiveness level in NHS resources. Nice, as an independent body that provides references to drug catalogue decision-making in the NHS, has statutory powers to determine whether drugs and medical technology enter the national drug reimbursement catalogue.Crizanlizumab (commodity name Adakveo) is a monoclonal antibody drug developed by Novart for P-selective, a cell adhesive protein found on the surface of blood vessel endoskines that plays a key role in multicellular interactions that can cause blood vessel blockages. The drug limits the interaction between endotrine cells, plate plates, red blood cells, and white blood cells by binding to P-selectives on the surface of the biopulation endotrine cells and plate plates. In November 2019, the drug was approved in the United States to prevent vascular obstruction in patients with sickle cell anemia. The FDA considers crizanlizumab to be a "first-in-class" drug, said to be the first and only targeted biologic agent to work by binding to P-selectives.In November 2020, the European Commission (EC) also approved Adakveo to prevent relapsed VOCs or pain crises in patients aged 16 and over with sickle cell disease as an additional therapy for hydroxyurea (including HC/HU) or as a monotherapy for patients with inappropriate or undertreated HC/HU. Its approval was based on data from phase 3 clinical SUSTAIN trials, which showed that Adakveo significantly reduced the average annual incidence of VOCs in sickle cell disease patients (1.63 to 2.98), equivalent to a 45 percent reduction, compared to placebo. But even if there are significant benefits, and the UK's drug regulator and health insurance are clearly more focused on balancing drug efficacy with long-term cost-effectiveness, Adakveo may need more discounts to break into the UK sickle cell disease market.
