Recently, Professor Zhao Wei's team from school of basic medicine of Shandong University has made a breakthrough in the research on the mechanism of innate immune activation. However, the interaction between redox homeostasis and DNA recognition signaling pathway, especially the role of lipid redox homeostasis in innate immune initiation is not clear.this study found that the lipid redox homeostasis maintained by glutathione peroxidase 4 (GPx4) is essential for sting activation.lipid peroxidation metabolite 4-HNE can promote the carbonylation of sting, and inhibit the palmitoylation and translocation from endoplasmic reticulum to Golgi.GPx4, as a key inhibitor of lipid peroxidation, promotes sting activation by maintaining intracellular lipid redox homeostasis.in this study, DNA virus HSV-1 infection model was used to reveal how the virus regulates lipid peroxidation, inhibits the host's antiviral immune response and realizes immune escape mechanism.this new mechanism of sting activation deepens the understanding of DNA recognition pathway and innate immune activation. At the same time, it provides a new strategy to regulate sting activation by regulating GPx4 activity and lipid peroxidation, which provides a new target for the prevention and treatment of diseases related to sting activation imbalance.Professor Zhao Wei's team has long been engaged in the study of viral infection and innate immunity, revealing the activation and regulation mechanism of innate immunity in the process of viral infection; relevant research results were published in NAT.
