    In recent years, the development of biocatalytic reactions has promoted the application of multifunctional enzyme systems in the field of biomedicine.

    However, compared with small molecule drugs, protein drugs are still facing many difficulties in clinical application due to their large size and easy inactivation, and the low protein loading efficiency of traditional drug carriers.

    Therefore, it is of great significance to design a functional carrier with high biological safety and excellent enzyme loading efficiency for protein drug delivery.

    The lower toxicity, good biocompatibility and higher drug loading efficiency of nanocarriers are the key factors to solve the problem of drug delivery in the treatment of diseases.

    Gallium-indium alloy (LM; gallium, 75%; indium, 25%; melting point: 15.
    7 ℃) has received more and more attention due to its good biocompatibility and low non-specific toxicity.

    At room temperature, liquid metal (LM) can react with acid to produce hydrogen, but it hardly reacts with water.

    As the temperature increases, LM will slowly react with water to produce rod-shaped gallium oxyhydroxide.

    What's more interesting is that the continuous heat treatment (temperature of 40 ℃ for 10 days) will cause the LM to be further etched, forming thorn-like structures at both ends, generating thorn-like nano-drums.

    The formation of thorn-like structure increases the surface roughness of LM, increases its specific surface area, and improves the adhesion of LM to biological macromolecules.

    Using the deformable characteristics of LM, the team of Professor Zhang Xianzheng of Wuhan University designed and synthesized a thorny cascade catalytic nanomaterial for photothermal and cascade catalytic treatment of tumors.

    Firstly, spherical LM nanoparticles were prepared by ice-water bath ultrasonic method, and then heat-treated for 15 days to obtain barbed drum-shaped LM.

    By testing the adsorption capacity of the barbed LM nanodrum and the spherical LM on different electrical proteins, it is confirmed that the barbed LM nanodrum has a better adsorption performance for protein than the spherical LM under the same conditions.

    Subsequently, amine oxidase (PAO) was immobilized on the surface of the barbed LM nano-drum (LMP), and the gallocatechin gallate (EGCG)-Fe3+ complex (LMPE) was wrapped to obtain the nano-catalytic material.

    In the acidic environment of the tumor, Fe3+ dissociates from the LMPE, and the exposed PAO will oxidize spermine (Spm) and spermidine (Spd) in the tumor tissue to generate H2O2 and aldehydes.

    The aldehydes produced will consume intracellular glutathione (GSH) and increase intracellular oxidative stress.
    At the same time, the generated H2O2 will be converted into highly active hydroxyl radicals (•OH) under the catalysis of Fe3+, which is harmful to tumor cells.
    Produce irreversible killing effect and realize highly effective cancer treatment.

    Combined with the excellent light-to-heat conversion performance of LM, under the guidance of multi-modal imaging, LMPE shows good tumor suppressive effects in mice both in vivo and in vitro.
