Background: osteosarcoma is the most common primary malignant bone tumor, and its poor prognosis and limitations of treatment methods determine that the development of new drugs for osteosarcoma is imminent.Objective: To investigate the anticancer effect of evodiamine (Evo) on human osteosarcoma cell lines 143B and MG63 in vitro, and to explore its potential mechanism.methods: human osteosarcoma cell lines 143B and MG63 were treated with a certain concentration gradient of Evo. MTT method, crystal violet method and colony forming test were used to detect the effects of Evo on the proliferation of the two cell lines.scratch test, Transwell test and matrix gel test were used to detect the migration and invasion ability of the two kinds of cells.the apoptosis of the two kinds of cells was detected by hochester 33258 staining.then the cell cycle changes and related protein expression levels of 143B were detected by flow cytometry and Western blot.luciferase reporter gene system was used to analyze the activation of Wnt / β - catenin signaling pathway in 143B cells, and Western blot was used to detect the expression of key factor β - Catenin and downstream target proteins in the signaling pathway.results: the results of MTT assay, crystal violet assay and colony formation assay showed that Evo inhibited cell proliferation in a dose-dependent manner.the results of cell migration and invasion test also showed that it had related inhibitory effect. Hoechst 33258 staining showed that Evo induced apoptosis in a dose-dependent manner.flow cytometry showed that 143B cells treated with Evo were arrested in G2 / M phase.in addition, the luciferase activity in the evo treated group was significantly lower than that in the untreated group, and the expression level of the corresponding pathway protein also proved that Evo played an anti-cancer role in osteosarcoma cells by inhibiting Wnt / β - catenin signaling pathway.conclusion: Evo can inhibit the proliferation, migration and invasion of human osteosarcoma cells 143B and MG63, and promote cell apoptosis in a dose-dependent manner in a certain period of time.
