Author: Ashley Introduction: New methods have been researched and developed for cancer treatment.
Recent studies have discovered the mechanism by which ERK5 protein kinase inhibitors destroy the proliferation of cancer cells and induce their death.
In the mechanism, ERK5 inhibits the activation of cytotoxic autophagy and triggers the death of cancer cells without affecting healthy cells
    .

Studies have found that autophagy regulation is a new treatment strategy for cancer: drugs that induce high levels of autophagy can cause the collapse of tumor cells and the activation of apoptotic cell death programs
    .

The synthesis of cellular protein occurs in the endoplasmic reticulum
    .

In response to different factors, such as lack of nutrients or oxygen, the endoplasmic reticulum becomes stressed.
This process can impair cell survival
    .

To solve this problem, a process called UPR (Unfolded Protein Response) was started to restore protein production and normal cells
    .

Among other strategies, UPR initiates autophagy, a biological process that allows cells to degrade and recycle defective components
    .

However, if the pressure is extreme or prolonged, UPR is not enough to restore protein production, UPR leads to cytotoxic autophagy and activates apoptosis (cell suicide)
    .

UPR occurs in all cells of the body, but has special relevance in tumor cells that exhibit high levels of endoplasmic reticulum stress
    .

UPR and autophagy enable tumor cells to better adapt to the environment and avoid the immune system
    .

Therefore, tumor cells are very sensitive to achieving autophagy levels that are toxic to the cells
    .

Therefore, autophagy regulation is a new treatment strategy for cancer: drugs that induce high levels of autophagy can cause the collapse of tumor cells and the activation of apoptotic cell death programs
    .

MAP kinase ERK5 protein controls the proliferation and survival of tumor cells and is a new therapeutic target for cancer treatment
    .

ERK5 inhibitors have shown curative effects in different cell and tumor models as monotherapy and in combination with chemotherapy
    .

However, the mechanism by which these inhibitors induce tumor cell death remains unclear
    .

A research team from the Wald Hebron Institute (VHIR) and the Autophagy University of Barcelona (UAB) led by José Miguel Lizcano revealed the mechanism by which ERK5 inhibitors cause cancer cell death.
The related study is entitled "ERK5 Inhibition Induces Autophagy-Mediated Cancer Cell Death by Activating ER Stress"
    .

Using human tumor cell cultures from pancreatic cancer, endometrial cancer, and cervical cancer, researchers found that ERK5 regulates tumor autophagy
    .

Therefore, ERK5 inhibitors activate endoplasmic reticulum stress and UPR (increased in tumor cells) to a level that exceeds their protective effect, activating a toxic form of autophagy and ultimately lead to tumor cell death (this process is called For cytotoxic autophagy)
    .

José Miguel Lizcano said: "ERK5 inhibitors make tumor cells sensitive to chemotherapy.
For this reason, our research has opened up a very promising path for improving chemotherapy and more effective strategies to deal with cancer
