Laboratory Guliang Yang, Haiyan Zhong and Shiming Li of Huanggang Normal College, etc., based on the rat RA model induced by bovine type II collagen (BIIC) Tangeretin, TAN) and 5-hydroxy-6, 7, 8, 3', 4'-methoxyflavone (5-hydroxy-6, 7, 8, 3', 4' pentamethoxyflavone, 5-HPMF) have a preventive effect on RA and its molecular mechanisms. After 5 weeks of INTERVENTION by TAN or 5-HPMF (TAN/5-HPMF), RA lesions and autophagy levels in the membrane tissue were significantly reduced, while ROS content and HO-1 expression levels were decreased. After TAN/5-HPMF feeding, the relative expression levels of p-AKT and p-mTOR were lowered. At the same time, the relative expression level of p62 in the TAN/5-HPMF treatment group was more than 2 times higher than that of the BIIC group. Immunofluorescent staining and Western blotting results further confirm that TAN/5-HPMF treatment reduces BIIC-induced conversion from LC3I to LC3II. Observations under a transmission electron microscope also showed a decrease in autophagy levels under TAN/5-HPMF intervention. These results show that TAN and 5-HPMF prevent the pathological process of BIIC-induced arthritis by inhibiting autophagy of membrane cells achieved by the ROS-AKT/mTOR signal axis.
