Carriers of the BRCA1 mutation have a higher risk of developing triple-negativebreast cancer(TNBC), a refractive disease that does not respond to current clinical targeted therapiesusing the Sleeping Beauty transposonsystem in Brca1 missing mice, the Deng early summer team at Xiamen University identified 169 predicted cancer drivers, including NOTCH1 as the primary candidate to accelerate TNBC by promoting epithelial interstitial transformation (EMT) and regulating cell cyclesactivation of NOTCH1 can suppress fissional fragmentation caused by BRCA1 deletion by activating the ATR-CHK1 signal path and restoring the S/G2 and G2/M cell cycle checkpointsconsistently, analysis of breast cancer tissue in humans showed that NOTCH1 was expressed highly in TNBCs, and that the activation form of NOTCH1 was positively correlated with the increase of phosphorylation of ATR, the researchers demonstrated that inhibition of NOTCH1-ATR-CHK1 works with cisplatin and can be used to kill TNBC in a coordinated way with target cell cycle checkpoints, DNA damage, and EMT, providing an effective clinical option for this deadly disease
