    Outline:Immune checkpoint proteins, including programmed cell death protein 1 (PD-1) and cytotoxic T lymphocyte-associated antigen 4 (CTLA-4), can bind to their respective receptors to promote immune escape by blocking the activation signal of immune cells
    .
    This immune evasion ability of tumor cells given by immune modification of immune checkpoints makes immune escape one of the important features of
    tumors.
    Immune checkpoint blocking (ICB) therapy targeting the above molecules inhibits key tumor signaling pathways through antibodies and specific small molecule inhibitors, and cracks the immune tolerance of tumor patients, which has shown excellent clinical benefits
    .
    Therefore, deciphering the role and regulatory mechanisms of these immune checkpoint molecules will provide key theoretical guidance
    for clinical treatment.
    In this review, we summarize current information about the function and regulatory mechanisms of these immune checkpoint molecules at multiple levels, including epigenetic regulation, transcriptional regulation, and post-translational modification
    .
    In addition, we summarize drugs that target various nodes in the regulatory pathway to further elucidate the clinical potential of newly identified immune checkpoint molecules, highlighting their potential implications
    for cancer diagnosis and immunotherapy.
