The global spread of the new crown virus poses a serious threat to human health and social economy
Recently, led by Professor Huang Ailong, the new crown virus emergency response team led by Professor Jin Aishun, and the key laboratory of medical molecular virology/BSL-3 laboratory of Fudan University, the Yang Haitao research group of Shanghai University of Science and Technology, and the Zhang Bo research group of Wuhan Institute of Virology Multidisciplinary cooperation, published the latest research results titled "Potent SARS-CoV-2 neutralizing antibodies with protective efficacy against newly emerged mutational variants" online in "Nature communications"
With the joint efforts of Professor Huang Ailong, Professor Jin Aishun and other researchers from Chongqing Medical University, more than 209 strains of RBD-specific antibodies have been obtained, and more than 100 strains of pseudovirus neutralizing antibodies have been identified, of which 3 have the strongest neutralizing activity Antibodies (58G6, 13G9 and 510A5) show super potent neutralizing ability against a variety of common pseudoviruses or live virus mutant strains, including B.
Cryo-EM structure analysis revealed that two of the antibodies (58G6 and 13G9) completely bound to the RBD in the "up" state on the S protein trimer; 58G6 bound to the S445-463 and S470-495 spatial structure regions of the RBD, and 13G9 only bound S470-495 spatial structure region, these structural analysis results explain the potential mechanism of our neutralizing antibody resistance to mutant strains, and also provide a theoretical basis for the later application of antibody drugs and the mechanism of mutant strain resistance to neutralizing antibody
To sum up, the research team of this project has discovered super-strong antibodies with broad-spectrum neutralization activity against multiple mutant strains of the new coronavirus.
The neutralizing antibody discovered by the research team was assisted by Wang Youchun's team from China Food and Drug Control Research Institute
After the outbreak of the new crown in early 2020, Professor Huang Ailong led the team to initiate the emergency response to the new crown virus as soon as possible, and the new crown virus neutralizing antibody drug research and development team quickly established an antibody screening technology platform
Postdoctoral fellows Li Tingting and Wang Yingming of Chongqing Medical University, assistant researcher Han Xiaojian, postdoctoral fellow Gu Chenjian of Fudan University, Nanjing University and Shanghai University of Science and Technology co-trained doctoral student Guo Hangtian, and researcher Zhang Huajun of Wuhan Institute of Virology are the co-first authors of this paper.
