Masahiro Sadakane, a professor of applied chemistry at Hiroshima University's Graduate School of Advanced Science and Engineering, said the resulting images are clear, but the process of acquiring and storing the required radioactive heavy metal solutions can be a complex hurdle for researche.
They published their findings May 12 in Scientific Repor.
"Observing virus morphology is essential in virology, and TEM is the most widely used technique because it enables direct visualization at the nanoscale, but currently it requires negative staining containing heavy elements," said Sadakane, corresponding author of the pap.
The current commercial alternative to radioactive uranyl acetate is a phosphotungstic acid called "keggin typ.
"We've been studying phosphotungstate compounds, and it was previously reported that the 'preyssler type' can be used as a negative staining reagent to visualize the fine structure of bacteria," Sadakane sa.
Preyssler-type molecules also include tungsten, oxygen, and phosphate, but they are structured around encapsulated positively charged ions, such as sodium or calci.
The researchers used preyssler-type phosphotungstates to stain and image three bacterial viruses (phages) that infect bacter.
"Our results show that preyssler-type phosphotungstates are good negative staining reagents for virus observation," Sadakane sa.
According to Sadakane, the researchers plan to build on their findings by developing a series of non-radioactive negative-stain reagents to look at other viruses, as well as small organic particles such as protei.
