From April 17th to 21st, the 2021 American Academy of Neurology (AAN2021) virtual meeting will be held online as scheduled.

A study by Wu David of the University of Minnesota and Zhejiang University of Traditional Chinese Medicine Huang Huijun, Qian Yu, Mao Yingying, etc.
, was announced in the form of a poster abstract at the 2021 American Academy of Neurology (AAN2021) annual meeting.

This study aims to evaluate whether there is a potential causal relationship between circulating linoleic acid (LA) levels and ischemic stroke (IS) through Mendelian randomization (MR).

Research Introduction LA is an essential fatty acid in the synthesis of eicosanoids.

Epidemiological studies have shown that there is an inverse association between circulating LA levels and IS.
However, whether this association is causal is still unclear.

The researchers obtained summary statistics on IS from the Megastroke Alliance, including 34,217 IS cases and 404,630 control cases of European descent.

In MR analysis, 17 single nucleotide polymorphisms (SNPs) related to circulating LA levels were used as instrumental variables (IVs), and the other two sets of SNPs were used for sensitivity analysis.

The inverse variance weighting method was used to assess the potential causality between circulating LA levels and IS.

Main findings: ➤For each increase of one standard deviation of the LA level predicted by the gene, the incidence of IS decreases by 2% (95%CI: 1%-3%).

➤Subgroup analysis showed that there was a significant causal relationship between LA level and aortic stroke (OR: 0.
    95; 95% CI: 0.
    92-0.
    98; P=3.
45×10-4), but with cardiogenic stroke (OR: 0.
    98; 95% CI: 0.
    96-1.
    00; P=0.
05), small vessel stroke (OR: 1.
    02; 95% CI: 0.
    99-1.
    05; P=0.
    11) had no significant causal relationship.

The sensitivity analysis using the other two sets of SNPs as IVs also yielded consistent results.

The study concluded that there is a causal relationship between circulating LA level and IS risk, especially with aortic stroke.

However, the potential mechanism of LA to prevent IS needs to be further studied.
