Background: Psoriasis is a common chronic inflammatory proliferative skin disease characterized by abnormal proliferation and differentiation of keratinocytes and immune T cells
psoriasis psoriasis
The Chinese medicine disease of psoriasis is called " white boil ", which is caused by multiple factors of loss of body nutrition and blood, internal blood heat, dryness and wind, and skin dystrophy
White scabbard white scabies nutrient- blood loss, blood heat inside, resolve dryness and generate wind, skin dystrophy, nutrient- blood loss, blood heat inside, change dryness and generate wind, skin dystrophy, clear heat, cool blood, detoxify , clear heat, cool blood, detoxify , nourish blood and nourish yin Moisturizing, nourishing blood, nourishing yin and moisturizing dryness
Yangxuejiedu Decoction (YJS) is a famous traditional Chinese medicine formula for the treatment of " blood dryness syndrome ".

Objective: To investigate the therapeutic effect of YJS on psoriasis model mice and its molecular mechanism
Methods: 50 BALB/c male mice were randomly divided into 5 groups (n=10): control group , IMQ group (imiquimod) , YJS+IMQ group , PES (Hsp70 inhibitor)+IMQ group , MTX group (Methotrexate) + IMQ group
Control group IMQ group (imiquimod) YJS+IMQ group PES (Hsp70 inhibitor)+IMQ group MTX (methotrexate)+IMQ group
According to the Clinical Psoriasis Area and Severity Index (PASI) , the severity of skin lesions was observed and calculated for 6 consecutive days (days 4-9); skin pathological changes were observed by HE staining ; CD4+ and CD8 were analyzed by immunohistochemistry and immunofluorescence + expression; ELISA was used to detect the levels of inflammatory factors TNF-α, IL-6, and IL-1β in each group; exosomes were extracted using a whole exosome isolation kit , and western blot was used to detect the levels of HSP70, CD63 and Calnexin proteins in exosomes.
Clinical Psoriasis Area and Severity Index (PASI) HE staining Immunohistochemistry and immunofluorescence analysis ELISA extraction of exosomes western blot
Results: YJS could effectively inhibit the hypertrophy, erythema and scale of psoriatic skin lesions , alleviate the pathological changes of skin lesions, and further reduce TNF-α, IL-6, IL-1β, IFN-γ, IL-17 and IL -23 production
Effectively inhibits hypertrophy, erythema and scaling of psoriatic-like skin lesions, inhibits HSP70 secretion, inhibits TLR4 activation and NF-κB p65 nuclear transfer
Conclusion: The therapeutic mechanism of YJS may be achieved by inhibiting the secretion of HSP70 from exosomes , preventing the activation of TLR4, and inhibiting the phosphorylation signaling pathway of NF-κB, further reducing the production of inflammatory cytokines and mediators
