    On May 30 , 2022 , Liu Tao's team from the State Key Laboratory of Natural and Biomimetic Drugs, School of Pharmacy, Peking University, together with Luo Xiaozhou's team from the Institute of Synthetic Biology, Shenzhen Institutes of Advanced Technology, Chinese Academy of Sciences, and Liu Xiaoyun's team from Peking University's School of Basic Medicine, have published an international journa.


    Post-translational modification of histones is one of the important research contents of epigenetics, and it is also an important target for drug development, such as the development of HDAC inhibito.


    Methanogens in nature have a set of orthogonal translation machines that can recognize amber codons ( TAG ) and encode pyrrolysines other than 20 classical amino acids into proteins through genetic codi.


    The genome-encoded wild-type histone H3 in the pM56 strain was replaced by the CTM histone H3 mutant, whose 56 -position non-canonical amino acids were identified by screening the optimized Saccharomyces cerevisiae GCE system ( PLRS/4xtRNA Leu.


    To study the effect of different acylation modifications of histones on DNA damage repair, the researchers treated the pM56 strain with DNA damage reagent.


    In this study, through interdisciplinary cooperation, based on the gene codon expansion system of Saccharomyces cerevisiae, exploratory site-directed co-translational modification was used as a research method for synthetic epigenetics, and another possible life form was created by artificial introducti.
