Alzheimer's disease (AD) and related dementias pose a growing challenge, imposing enormous costs on personal, health and social care services

Furthermore, longitudinal studies have shown that most cognitively normal amyloid-positive individuals never develop clinical dementia

To this end, experts from the Department of Epidemiology and Public Health at UCL, UK, examined the association between 4,953 plasma proteins and the risk of cognitive decline and dementia in two cohort studies followed for 20 years.

In the Whitehall II prospective cohort study, proteins were measured using the SOMAscan technique

246 of the 4953 proteins were associated with an increased rate of cognitive decline

In analyses adjusted for age, sex, and ethnicity/ethnicity, 1SD higher levels of each of the 15 proteins were associated with increased risk ranging from 8% to 64%

This study identified several plasma proteins associated with cognitive decline and long-term risk of dementia in people without dementia
