Purpose : Myofiber necrosis is an important pathological feature of idiopathic inflammatory myopathies ( IIMs ), and the molecular mechanisms are largely unkno.

Methods Western blotting and immunohistochemical analysis were performed to examine the expression of receptor-interacting protein 3 (RIP-3) and MLKL proteins , as well as necroptosis -related injury - related molecules in 26 IIM patients and 4 healthy contro.
Results Compared with healthy controls, the expression of RIP-3 and MLKL proteins and their phosphorylated forms were significantly increased in muscle tissue of IIM patient.
Conclusions These findings suggest that hyperactivated necroptosis leads to muscle damage in IIM , and suggest that necroptosis inhibitors may represent novel therapeutic targets for the treatment of II.
