"Our study shows that we can use germ-free animals as hosts to generate germ cells from other animal species," said senior author Ori Bar-Nur, a stem cell biologist at ETH Zuri.
Pluripotent stem cells (PSCs) provide a powerful tool for biomedical research, but generating gametes in the form of eggs or sperm cells from PSCs is an extremely challenging endeav.
To test this idea, the researchers injected rat PSCs into mouse embryos to create mouse-rat chimer.
"We were surprised by the relative simplicity of the two species, and we could mix them to produce viable mouse-rat chimer.
Although the researchers were able to generate rat sperm cells that were morphologically indistinguishable from normal rat sperm cells, the cells were immobile and the rate of fertilization of rat eggs was higher than that of rat sperm cells produced in rats significantly reduc.
Next, the researchers will try to generate living animals from mouse sperm cells created from mouse-rat chimer.
A longer-term plan is to apply the technology to gamete production in endangered rodents to support animal species conservation effor.
