With the advancement of molecular biology technology, especially DNA sequencing technology, people's understanding of cancer cell genomes has become comprehensive, detailed and in-depth, and they have begun to continuously revise their cognition of the correlation between "cancer occurrence and gene mutation"
Recently, the scientific research team of the Van Andel Institute in the United States discovered that the main factor that promotes the generation of genetic mutations in melanoma comes from the chemical transformation of DNA driven by sunlight, rather than DNA replication errors
Related research was published in Science Advances with the title The major mechanism of melanoma mutations is based on deamination of cytosine in pyrimidine dimers as determined by circle damage sequencing
Melanoma is a tumor produced by the precipitation of melanin in the skin or other organs.
As we all know, melanoma has the most DNA mutations of all cancers.
Previous studies have found that UVB radiation can induce various forms of DNA damage, resulting in the main products that induce skin cancer, "cyclobutane pyrimidine dimer (CPD)" and "pyrimidine (6-4) pyrimidinone dimer (6).
The sequence specificity of cytosine deaminated CPD and its relationship with melanoma mutations
However, in this latest study, researchers have discovered a new mechanism for cancer treatment mutations, that is, when UVB radiation hits the base sequence in DNA, it will seriously affect its stability and cause "pre-mutation".
CPD deamination and potential mutagenesis
But it is worth noting that this kind of mutation may not induce cell cancer immediately.
Therefore, in order to effectively treat and prevent cancer, it is urgent to find out the source of mutations, and a "circular damage sequencing method (CD-seq)" developed by researchers seems to make it possible to fight cancer.
The outline of CD-seq
The study pointed out that the sequencing method can "destroy" the DNA damage, induce it into a circle, and then amplify it based on PCR technology.
