*Background: it is reported that nudt21 is an RNA binding protein and plays an important role in the regulation of a variety of biological reactions.the expression of nudt21 may be a new marker for the detection of breast cancer.Objective: This study is to explore the function and clinical significance of nudt21 in breast cancer.methods: the protein expression of nudt21 was detected by immunohistochemistry (IHC) in 100 paraffin embedded, archived breast cancer and 100 benign breast tissue samples.then the correlation between the expression of nudt21 and clinicopathological features and prognosis of breast cancer patients was analyzed.in addition, the function of nudt21 in breast cancer cell lines was detected by MTT, colony formation and Transwell assay.finally, the proteins directly interacting with nudt21 were identified by mass spectrometry and Western blotting.results: immunohistochemistry (IHC) analysis showed that the expression of nudt21 in breast cancer was significantly lower than that in benign breast disease tissues.correlation analysis showed that the low expression of nudt21 was positively correlated with tumor size, lymph node metastasis and TNM stage.similarly, Kaplan Meier survival curves showed that patients with lower nudt21 expression had shorter overall survival (OS) and relapse free survival (RFs) than patients with higher nudt21 expression. In addition, knockdown of nudt21 enhanced cell proliferation, migration, invasion and epithelial mesenchymal transition (EMT).similarly, the overexpression of nudt21 inhibited cell proliferation, migration, invasion and EMT.in addition, nudt21 directly interacts with cpsf6 and negatively regulates its expression.cpsf6 knockout can reverse the migration and invasion of cancer cells induced by nudt21 expression.conclusion: nudt21 may inhibit the proliferation and invasion of breast cancer cells through nudt21 / cpsf6 signaling pathway.key words: nudt21, EMT, cpsf6, breast cancer
