Recently, Yang Litao's research group from Zhang Dabing's School of Life Science and Technology, Shanghai Jiao Tong University published an online publication entitled "LIFE-Seq: A universal Large Integrated DNA Fragment Enrichment Sequencing strategy for deciphering the transgene integration of The research paper of "genetically modified organisms" has established for the first time a large fragment capture and sequencing technology suitable for the analysis of molecular features such as integration sites and flanking sequences of exogenous DNA of various transgenic organisms

The molecular features of genetically modified organisms (GMOs) mainly include information such as the integration site of exogenous DNA in the recipient genome, the complete insertion sequence and structure, flanking sequence and copy number

The study firstly screened the main commonly used transgenic elements, exogenous genes, standard genes in species and other construction sequence data sets from the genetically modified organism detection method database (GMDD) established by the team in the early stage, and designed a set of high-coverage oligonucleotides.

    Figure 1.

In the experiment, six transformants of transgenic corn, soybean, rice and rapeseed in commercial application were used as experimental materials, test samples with different transgenic content and non-transgenic control samples were designed, and the feasibility and accuracy of the LIFE-Seq method were systematically tested

    Figure 2.

The results showed that LIFE-Seq successfully analyzed the molecular features of exogenous DNA integration in a variety of transgenic crop transformants clearly and accurately

Yang Litao's research group has made a series of progress in the identification and detection of GMO molecular features in recent years (J Agric Food Chem, 2021, 69, 1705-1713; ACS Agric.
