January 29, the Chinese Academy of Sciences Institute of Biophysics Biometrics National Key Laboratory Rao Zi and the research team, in cooperation with the Air Force Military Medical University Xu Zhikai task force, published a research paper on
online entitled "Revealing the structural basis and mesothetic mechanism of high school and active antibodies of encephalitis B virus". The work reported the frozen electroscopic structure of JEV of encephalitis B virus and two therapeutic antibodies 2F2 and 2H4 compounds respectively, and expounded the function and mechanism of neutral antibodies of the two strains. This is another important research result of Rao Zi and his team after analyzing the three-dimensional fine structure of all-particles of the type B encephalitis virus (JEV).
mortality rate was higher in patients with encephalitis B, and survivors were susceptible to neurological sequelae. Therefore, the development of specific drugs and new vaccines for encephalitis B virus is of great importance to the prevention and treatment of the epidemic. 2F2 and 2H4 are the high school and active antibodies screened by the Department of Microbiology of the Fourth Military Medical University 30 years ago, which were used in patients' clinical treatment during the special period of frequent outbreaks of encephalitis B at that time, and showed good safety and therapeutic effect.
In the study, the researchers first found that the neutral antibodies 2F2 and 2H4 interacted only with the type B encephalitis virus, not with dengue, Zika, and yellow viruses of the same genus, and that both antibodies showed a high affinity. Through cell-level meso-experiments, it was found that both antibodies have excellent between the two antibodies. Animal-level conservation experiments have found that the two antibodies completely remove the encephalitis B virus from the brains of infected mice and completely cure infected mice.
the composite structures of 4.7 s JEV-2F2 and 4.6 s JEV-2H4 by using a frozen electroscope. Through structural analysis, it is found that the antibodies with higher neutrality and activity in the yellow virus genus are combined in the four-stage structure of E protein, and these antibodies are virus-specific neutral antibodies. Therefore, according to the above characteristics, the specific new vaccine for the virus can be designed. By further studying the mechanisms of action of the two neutral antibodies, the researchers found that the two antibodies not only blocked the binding of viruses and cells, but also prevented the fusion of viruses and cells under acidic conditions, and played a leading role.
the research was supported by the National Natural Science Foundation of China, the Ministry of Science and Technology 973 Project, the Chinese Academy of Sciences pilot special project, and the support of Qin Chengfeng, a researcher at the Institute of Military Medicine, and Liu Shanhuai, a researcher at Ohio State University. Wang Xiangxi, a researcher at the Institute of Biophysics of the Chinese Academy of Sciences, Xu Zhikai, a professor at the Air Force Military Medical University, and Rao Zi, a member of the Chinese Academy of Sciences, are co-authors of this paper, as are Qiu Xiaodi, a team doctoral student, and Lei Yingfeng, an associate professor at the Air Force Military Medical University.
