A clear understanding of the neural correlates of depression in Parkinson's disease remains elusive, with functional, structural, and nuclear imaging studies yielding extensive findin.
It occupies a key position in the basal ganglia and cortical networks and is implicated in the pathogenesis of both PD and mood disorde.
Although white matter microstructural changes and noradrenergic dysregulation in medial thalamic regions have been identified, these studies were unable to examine changes within individual thalamic subnuclei, limiting anatomical precisi.
Structural changes can be found in areas with altered functional connectivity following antidepressant treatment, and antidepressants may reduce gray matter atrophy in affected areas through neuroprotective, neurogenic, and neuromodulatory effec.
Until recently, analyzing the morphology of thalamic subnuclei and the specific neural connections of individual nuclei has proven problematic because it requires manual segmentation of thalamic subnuclei, which can be laborious and error-pro.
Hereby,.
They hypothesized that depression was associated with smaller volume and altered white matter structure within the thalamic subnucl.
They performed structure- and diffusion-weighted imaging (DWI) in 76 PD participants to assess the relationship between PD depression and changes in gray and white matter thalamic subnuclea.
Fixel-based analysis was used to calculate the average fiber cross-section (FC) of the white matter tracts attached to each subnucle.

They found that PD depression scores were associated with lower right anterior vertebral body (PuA) subnuclear volu.
After follow-up, depression scores were associated with reductions in nearly all large structures in white matter tracts that were connected to thalamic subnucl.
The significance of this study lies in the discovery that right PuA is a relevant neural structure in PD depression, and future work should evaluate its potential as a therapeutic target for PD depressio.
