    Introduction: Tumors often exploit immunosuppressive factors in their tumor microenvironment to impair T cell function, which in turn leads to T cell depletion, resulting in ineffective immunotherapies such as CAR-T cell therapy


    On August 24, scientists from the University of California, USA published a study "RASA2 ablation in T cells boosts antigen sensitivity and long-term function" in "Nature".

    Adoptive T cell therapy (CAR-T cell therapy) has made major breakthroughs in hematological tumors, and has also made great progress in solid tumors in recent years

    In order to screen for regulators that can counteract the suppressive effect of T cells, the researchers screened through the genome-wide CRISPR knockout library.

    To further determine the effect of RASA2 on T cells, the researchers analyzed the effect of RASA2 knockout on T cell activation, antigen sensitivity and effector function.

    The poor durability of CAR-T cell therapy has always been a major bottleneck in the application of this treatment in solid tumors.
