DNA errors stress cells, causing dysfunction
Inside brain cells, errors in DNA accumulate as we age
A new study by researchers at Brigham and Women's Hospital and Boston Children's Hospital finds that people with Alzheimer's have more somatic mutations in their brain cells than those without Alzheimer's.
"As we age, neurons accumulate somatic mutations
The team performed single-cell whole-genome sequencing of 319 hippocampal and prefrontal cortical neurons in AD patients or non-AD patients to determine the link between the number and type of somatic mutations and AD
Ultimately, the discovery that DNA alterations accumulate in AD neurons provides researchers with a window into the molecular and cellular events in AD pathogenesis
The authors acknowledge two major study limitations
"In the future, we are eager to elucidate how the observed mutations in AD neurons lead to neuronal cell death and work to help discover new treatments targeting these pathways," Miller said
