    Semantic variant primary aphdrenapath (svPPA) is a neurodegenerative disease characterized by loss of conceptual knowledge, usually characterized by difficulty finding words, impaired understanding of individual words, and atrophy of the temporal lobe.
    it is a type of frontal temporal lobe dementia (FTD) that is usually associated with TDP-43 C pathology.
    that although much progress has been made in understanding the cognitive and biological properties of svPPA over the past 20 years, little research has been done on fluid biomarkers.
    In this study, this paper examines the role of plasma glial fibrous acidic proteins (GFAPs) and neural filament light chains (NfL) as biomarkers of ascental glial cell activity and neurodegeneration in svPPA, as well as their relationship to the svPPA disease severity.
    blood plasma samples were collected from 64 consecutive subjects in the FTD queue study at University College London: 28 subjects who met the svPPA1 diagnostic criteria and 36 healthy controls.
    there was no statistical difference in age or sex between the two groups (t-test, p-0.934, Fisher-accurate test, p-1.000).
    participants received standardized clinical and cognitive assessments (Table 1), which included two measurement methods of semantic knowledge, the Webster Mental Strength Table (WASI) vocabulary quiz and the UK picture vocabulary scale.
    the brain was 3D T1-weighted MRI through Siemens Trio or Prisma 3T scanners, and 55 scans were performed to calculate the volume of grayscale in the temporal lobe (using the auto-segmentation technique described earlier to calculate the sum of the left and right hemispheres).
    GFAP and NfL were measured using the Newology 4-Plex A kit.
    consistent with clinical diagnosis, the svPPA group was significantly impaired on two semantic knowledge tests (WASI glossary, z=6.20, p<0.001; BPVS, z=6.35, p<0.001), and the lower volume of the temporal lobe (t=12.79, p<0.01).
    GFAP and NfL water levels were significantly related in the svPPA group (rho=0.53, p=0.004) and the control group (rho=0.51, p=0.002).
    in the svPPA group, NfL increase is independent of the degree of semantic damage (NfL:WASI glossary, rho=0.55, p=0.004; BPVS, rho=0.56, p=0.003; GFAP: WASI glossary, rho=0.22, p=0.291; BPVS, rho=0.26, p=0.204).
    NfL and GFAP concentrations were negatively corred with temporal lobe volume (NfL, rho=0.47, p=0.033; GFAP, rho=0.58, p=0.006).
    NfL is more widespread than GFAP, both GFAP and NfL can identify the svPPA's disease severity.
    with other neurodegenerative diseases, and the decline in their levels is likely to prove to be effective in future clinical trials.
