Isolation of cfDNA from serum samples was performed as previously described and the DNA copies of psoriasis-associated autoantigens were measured using droplet digital polymerase chain reaction (ddPCR)

First, we checked for the presence of cfDNA for psoriasis-related autoantigens
.
DdPCR showed that PLA2G4D DNA could be detected in normal people and patients with psoriasis (Figure 1a), but LL-37 DNA was not detected (data not shown).
The number of circulating PLA2G4D DNA copies in patients with psoriasis was significantly higher than that in healthy controls.
Secondly, we analyzed the relationship between circulating PLA2G4D DNA copy number and clinical manifestations of psoriasis (age, gender, psoriasis area and severity index (PASI) score, presence or absence of joint involvement), and found that circulating PLA2G4D DNA level is related to Age and gender are irrelevant
.
However, we did observe a weak positive correlation between circulating PLA2G4D DNA levels and PASI scores (Figure 1d)

PLA2G4D is a psoriasis antigen that is overexpressed in psoriatic skin lesions (keratinocytes, dendritic cells, macrophages and T cells)

PLA2G4D (as a lipid antigen) specific CD1a reactive T cells produce large amounts of IL-22 and IL-17A, and these molecular mechanisms induce psoriasis inflammation
