Alkaline Phosphatase (AKP/ALP) Activity Assay Kit
Instructions for Spectrophotometry 50 tubes/24 samples
Instructions for Spectrophotometry 50 tubes/24 samples
Note: Before the formal determination, select 2-3 samples with large expected differences for pre-determination
.
Determination meaning: Determination principle: Self-provided instruments and supplies Alkaline phosphatase (AKP/ALP) activity determination kit instructions
Reagent composition and preparation: crude enzyme liquid extraction:
- Tissue: According to the ratio of tissue mass (g):reagent one volume (mL) to 1:5~10 (it is recommended to weigh about 0.
1g of tissue and add 1mL of reagent one), homogenize in ice bath, centrifuge at 8000g at 4°C for 10min, Take the supernatant for testing
. - Bacteria or cells: According to the number of bacteria or cells (10 4 ): the ratio of reagent 1 volume (mL) is 500~1000:1 (it is recommended to add 1 mL of reagent 1 to 5 million cells), and ultrasonically break cells in ice bath (power 300w, ultrasonic 3 seconds, the interval is 7 seconds, the total time is 3 minutes); then 8000g, 4 ℃, centrifuge for 10 minutes, take the supernatant and put it on ice for testing
. - Blood can be measured directly or after appropriate dilution
.
Assay steps: AKP/ALP activity calculation:
- Calculation of AKP/ALP activity in blood
- Calculation of AKP/ALP activity in tissues, bacteria or cells
