Cyclic nucleotide phosphodiesterases regulate cellular levels of small molecule second messengers that control important biological processes in all kingdoms of life. Here, we describe the use of thin layer chromatography to analyze cellular extracts or purified proteins for cyclic nucleotide phosphodiesterase activity.
