Gastric cancer (GC) is the fifth most common cancer type in the world and the third leading cause of cancer-related death
The invasion and metastasis of gastric cancer is a difficult problem in the current clinical treatment
Recently, researchers from the Department of Gastroenterology, Xinqiao Hospital, Third Military Medical University, published an article entitled "Demethylase ALKBH5 suppresses invasion of gastric cancer via PKMYT1 m6A modification" in the journal Molecular Cancer
The researchers first performed methylated RNA immunoprecipitation sequencing (MERIP-SEQ) on gastric cancer tissues
This study found that the expression of ALKBH5 was decreased in gastric cancer tissue and was associated with clinical distant metastasis and lymph node metastasis
We also found that upregulation of PKMYT1 expression was caused by ALKBH5 knockout or mutation of demethylase activity, suggesting that ALKBH5 regulates PKMYT1 expression in an m6a-dependent manner
In conclusion, this study identified ALKBH5 as a tumor suppressor gene for gastric cancer metastasis, and this role was dependent on the demethylase activity of ALKBH5
