Lung cancer is the highest incidence rate and mortality rate in China and even worldwide.in China, there are more than 700000 lung cancer cases and more than 600000 deaths every year in China.lung adenocarcinoma is the main pathological type of non-small cell lung cancer, accounting for about half of all lung cancer.compared with other pathological types of lung cancer, the proportion of non-smokers in lung adenocarcinoma is significantly higher, and the pathogenesis is complex.early detection, early diagnosis and early treatment of lung adenocarcinoma are of great significance for improving the prognosis of patients.protein is the direct executor of all life functions, and almost the target of all drugs.the systematic study of all protein characteristics in biological samples, such as cells and tissues, has gradually become one of the core contents of biomedical research.therefore, it is of great scientific significance to draw a panoramic protein molecular map of lung adenocarcinoma population for in-depth understanding of the pathological mechanism of lung adenocarcinoma, the discovery of biomarkers for disease diagnosis and drug treatment targets, as well as the realization of more accurate molecular typing of lung adenocarcinoma and the formulation of treatment plans.on July 9, 2020, cell, an international top academic journal, officially published a large-scale clinical lung adenocarcinoma proteome sketch drawing work completed by Chinese scientists.in this study, the research team of Tan Minjia, Shanghai Institute of medicine, Chinese Academy of Sciences, together with Academician he Fuchu and Wang Yi from Academy of military medicine, National Center for Protein Science (Beijing), National Cancer Center / Chinese Academy of Medical Sciences Cancer Hospital Cheng Shujun and Xiao Ting, and Professor Li Jing of Shanghai Jiaotong University For the first time in the world, the team has carried out a large-scale, high-throughput and systematic panoramic proteomics study on lung adenocarcinoma.in this study, protein expression profiles and phosphorylation post-translational modification profiles of 103 patients with lung adenocarcinoma and paracancerous tissues were analyzed. A total of 11119 protein products and 22 protein products were identified, At the same time, we integrated clinical information and genomic data analysis to construct a proteome based molecular map of lung adenocarcinoma.for the first time, this study systematically described the molecular map of lung adenocarcinoma from the protein level system, and found the molecular characteristics closely related to the prognosis of patients, especially the protein molecular characteristics of two major genes (TP53 and EGFR) mutation in Chinese population.the researchers further used the proteomic expression profile data to classify lung adenocarcinoma into three proteomic subtypes (type I, II and III).among them, type I is closely related to cell metabolism and tumor microenvironment. Clinical information shows that this type is mainly early clinical population, and has the best prognosis; type III is closely related to cell homeostasis and proliferation, which is mainly in the middle and late clinical stage, showing low tumor differentiation, high gene mutation load and the worst prognosis; type II is type I and type III In the transitional state, the prognosis of this group is between type I and type III.combined with the data analysis of phosphorylation modification spectrum, this study also revealed the characteristics of kinase activity among different subtype populations, providing a basis for more accurate classification and treatment of lung adenocarcinoma.in order to further explore the potential application value of the research work in the clinical diagnosis and treatment of lung adenocarcinoma, 27 potential prognostic markers of lung adenocarcinoma with serological detection value and several potential drug targets for lung adenocarcinoma and its specific mutation subtypes were screened.the protein concentration of HSP 90beta is closely related to the poor prognosis of lung adenocarcinoma.these results provide important scientific clues and theoretical support for the pathological mechanism analysis, accurate diagnosis and treatment of lung adenocarcinoma.this work is the first large-scale and systematic construction of the protein panorama and molecular subtype characteristics of lung adenocarcinoma, revealing the molecular characteristics and biomarkers of prognosis and diagnosis and treatment of lung adenocarcinoma in China, which provides important resources and clues for the precise medical treatment of lung adenocarcinoma.this work is another major achievement of "Chinese proteomics program" following the work of liver cancer and gastric cancer, and another major breakthrough of "proteomics driven precision medicine" led by Chinese scientists.
