Introduction: Tumor metastasis is a malignant process influenced by many factors, among which the influence of tumor microenvironment (TME) is huge
On August 30, Zhu Di, a national youth special expert of Fudan University, and Professor Xu Jianmin of the affiliated Zhongshan Hospital jointly published a study "Tumor-infiltrated activated B cells suppress liver metastasis of colorectal cancers" in "Cell Reports", which found that tumor infiltration Activated B cells can inhibit liver metastasis of colorectal cancer
Doi: 10.
Research Background
01
Colorectal cancer (CRC) ranks third in the global incidence.
research process
02
To elucidate the immune cell profile of colorectal cancer liver metastases, the researchers analyzed samples from patients who underwent surgical resection of the primary tumor and divided them into four groups: (1) left-sided colorectal cancer liver metastases; (2) Right colorectal cancer liver metastasis group; (3) left colorectal cancer without liver metastasis group; (4) right colorectal cancer without liver metastasis group, and it was found that whether or not liver metastasis affects the immune cell population of colorectal cancer Activated B cells were significantly reduced in liver metastases in particular (Figure 1)
Figure 1.
To further evaluate the role of B cells in colorectal cancer metastasis, the researchers performed cluster analysis on B cells and found that patients with more B cells had a significantly longer survival time (Figure 2), and found that the left colorectal cancer liver Significant differences were found between B cells in metastases and right-sided colorectal liver metastases, and Wnt pathway signaling was closely related to B cell development
Figure 2.
In addition, the tumor infiltration rate of activated B cells was significantly reduced in right-sided colorectal cancer with liver metastases compared with right-sided colorectal cancer without metastasis, and the increased tumor infiltration rate of activated B cells inhibited the colorectal cancer mouse model Liver metastases in colorectal cancer, and the expression of activated B-cell markers was negatively correlated with the rate of liver metastases in colorectal cancer (Figure 3)
Figure 3.
Significance
03
The 5-year survival rate of colorectal cancer patients with liver metastasis is extremely low, and the role of B cells in colorectal cancer liver metastasis is still unclear
