Tumor metastasis is one of the ten characteristics of cancer. According to statistics, more than 90% of cancer deaths are closely related to tumor metastasis [1,2].more and more studies have pointed out that tumor microenvironment, especially various cytokines, plays an important role in the malignant development of tumors [3,4].therefore, it is one of the key problems to develop effective biological agents for the prevention and treatment of tumor metastasis with less toxic and side effects by using the latest progress of medical biology at home and abroad, combined with the latest science and technology of various disciplines.recently, Professor He Qingyu, researcher Li Bin and associate researcher Xu Wenwen, Key Laboratory of tumor molecular biology, Ministry of education of Jinan University, published the title "direct targeting of CREB1 with imperatorin inhibitors TGF β 2-erk signaling to suppress esophageal cancer" In this paper, we analyzed the expression of TGF β 2 in clinical samples based on public database and clinicopathological data. It was the first time to point out that TGF β 2 was highly expressed in esophageal cancer tissues and was negatively correlated with the survival rate of patients.innovatively using high-throughput drug screening platform, we found that imperatorin can significantly inhibit the invasion of esophageal cancer and TGF β 2 signaling pathway.this study, combined with a variety of cell models in vivo and in vitro, indicated that imperatorin can effectively inhibit the invasion and metastasis of esophageal cancer.in order to explore the molecular mechanism of imperatorin inhibiting invasion and metastasis, the research group used quantitative proteomics to compare the protein expression profiles between the esophageal cancer cells treated with imperatorin and the control group, and then bioinformatics analysis was carried out to find the related downstream pathways.the data suggest that imperatorin may exert its function by affecting TGF β 2-erk pathway.subsequently, through a series of molecular dynamics, molecular biology, clinical pathological analysis and other experiments, it was found that imperatorin can inhibit the phosphorylation of CREB1 and the transcription regulation of TGF β 2 through the direct binding of CREB1 protein, thus reducing the expression of TGF β 2, blocking the ERK signal pathway, and playing an inhibitory role in the invasion and metastasis of esophageal cancer.the research group studied the inhibitory effect of imperatorin on the invasion and metastasis of esophageal cancer from three aspects of biological function, clinical significance and molecular regulation mechanism. The research results provide new clues for the molecular markers of prognosis and drug selection in non-invasive diagnosis of esophageal cancer.Huang Zhihao, associate researcher and master of Jinan University of Sciences, is the co first author of this paper.
